Note that the only thing you can do with jutsu cards at this point is charge chakra.

Also note that at any point in the game you can click enlarge to view a full sized card.

1. Play a ninja card

To do this, click on "Put in play". You will not be able to play the ninja if you do not meet the requirements to play it, or if you have already played a ninja card. However, certain mission cards or ninja effects can allow you to play another ninja.

Your ninja may require a discard cost, which will bring up a discard dialog, asking to discard from your hand the required amount of cards that are the same element as the ninja you are trying to play. Keep in mind you can tell the element of the card by looking at the background colour

2. Charge chakra

Doing this will send the card to the chakra area.

Chakra breakdown

The chakra breakdown shows how much of each type of chakra is in the chakra area. Both you and your opponent will be able to see your chakra breakdown. Click anywhere in the chakra breakdown to see what cards are in the chakra area.

3. Play a mission card

If you meet the requirements to play this card i.e. entrance cost, you can attempt to play the mission card.

If the mission card requires a target, the context menu will say so, be it a team or an individual ninja as the target, just click on the team or the individual ninja respectively.

Similar to playing a ninja, if the mission card has a discard cost, the discard dialog will appear.

In the very first screen shot, you will notice that the mission card being selected to put in play is "Noodle Shop". This mission card allows you to heal an injured ninja in your village. Below will show the results of successfully playing this mission card.

Playing the mission card Noodle Shop on this injured ninja will heal him; going from having a red tinge to displaying normally
Mission outcome

As you can see, the ninja has a red tinge. This signifies that it is injured. Playing Noodle Shop on this injured ninja will heal it, and the results are on the right side. The ninja loses the red tinge, signifying it is healthy again.
